As a set painter for Columbia Pictures, McGavin started his career. Following roles in David Lean’s Summertime and Otto Preminger’s The Man with the Golden Arm, he played the lead in the Broadway productions of My Three Angels and The Rainmaker in 1954.

Darren McGavin appeared in Riverboat (1959–1961), Kolchak: The Night Stalker, and Mickey Spillane’s Mike Hammer on television between 1958 and 1959.

Darren McGavin received a Primetime Emmy nomination for Outstanding Guest Actor in a Comedy Series for his recurring part on the sitcom Murphy Brown.
