Stefan de Vrij is the youngest son of Jan de Vrij, who played for local club VV Spirit. Jan de Vrij is a former football player. Stefan de Vrij began his career at his local club VV Spirit and debuted as a professional for Feyenoord aged 17.

Stefan de Vrij was born on February 5, 1992, in Ouderkerk aan den IJssel, Netherlands. On 17 July 2009, Stefan de Vrij signed his first professional contract with Feyenoord until summer 2012.

On 6 December 2009, Stefan de Vrij made his Eredivisie debut in the home match against FC Groningen (3–1), when he replaced Denny Landzaat in the 89th minute.
Stefan de Vrij’s second season with Lazio began with the 2015 Supercoppa Italiana, playing the full 90 minutes of a 2–0 loss to Juventus in Shanghai.
Stefan de Vrij scored his first goal for Lazio in Serie A on 11 September 2016 in a 1–1 away draw to Chievo. On 9 November 2019, in the 2019–20 season, Stefan de Vrij made his 50th appearance in all competitions for Inter in a 2–1 league win over Verona at San Siro.
