Born in Greenwich, Connecticut, Close majored in theater and anthropology at the College of William & Mary.
She began her professional career on the stage in 1974 with Love for Love. While in Broadway, she appeared in productions of Barnum in 1980 and The Real Thing in 1983, winning the Tony Award for Best Actress in a Play for the latter.

Eventually, she was given a callback and hired for one season to do three plays at the Helen Hayes Theatre, one of those plays being Love for Love directed by Hal Prince.

She made her television debut in 1975 with a small role in the anthology series Great Performances.
In 1979, she filmed the television movies Orphan Train and Too Far to Go. The latter film included Blythe Danner and Michael Moriarty in the cast, and Close played Moriarty’s lover.

Glenn has appeared in a number of movies including but not limited to comedy drama The Paper (1994), Tim Burton’s alien invasion satire Mars Attacks! (1996), Air Force One (1997), where she played the trustworthy vice president to Harrison Ford’s president; Paradise Road (1997) as a choir conductor of the women imprisoned by the Japanese in World War II.
In 1999, Glenn provided the voice of Kala in Disney’s animated film Tarzan. She later went on to receive great reviews for her comedic role as Camille Dixon in Cookie’s Fortune (1999).
